目的

检测白细胞介素(IL)-17 在口腔扁平苔藓(OLP)病损局部组织的表达情况,初步探索Th17 细胞在OLP 发病过程中的作用。

方法

26 例OLP 活检组织和6 例健康颊黏膜组织制成匀浆,ELISA 测定IL-17 浓度,应用BCA 总蛋白检测方法测定总蛋白浓度,根据IL-17 相对含量(pg/mg)=细胞因子(pg/ml)/总蛋白(mg/ml),计算IL-17 相对含量;应用免疫组化方法检测组织中IL-17 表达的组织分布。

结果

在所有标本中均能检测出IL-17 的表达,对IL-17 相对含量结果进行非参数秩和检验,与对照组相比差异无统计学意义,IL-17 阳性染色主要在固有层呈散在分布, 在表皮不完整的糜烂处及上皮下疱处, 阳性细胞明显增多,上皮内极少见阳性染色。 高倍镜下IL-17 阳性染色主要分布在胞浆,部分细胞的胞外间隙也可见弱阳性染色。

结论

IL-17 在OLP 病损组织中均有表达,但与对照组相比,其表达水平差异无统计学意义;IL-17 在OLP 中的作用仍需进一步探讨。

Objective

To detect IL-17 expression in lesions of oral lichen planus.

Methods

Tissue homogenate liquids were obtained from 26 OLP biopsy specimens and 6 controls. The expression of IL-17 was quantitated by ELISA and the total protein concentration were detected by BCA total protein detection according to the manufacturer's instructions. The relative amount of IL-17 was calculated as the ratio of cytokine concentration to total protein. The expression and location of IL-17 was detected by immunohistochemistry.

Results

IL-17 could be detected in both experimental group and control group. The relative amount of IL-17 ranged widly, and the data did not normally distributed. IL-17 was detected at extremely high levels in some OLP specimens, however, when compared with that in normal buccal mucosal tissues, no significant difference was found. The IL-17 positive cells were found dispersedly in submucosa lamina propria of OLP, and more could be observed in erosive and vesicle lesions of OLP. The IL-17 located in cytoplasm predominantly, whereas there was some weakly positive staining in extracellular space.

Conclusions

Our results indicate that IL-17 positive cells distributed dispersedly in submucosa lamina propria of OLP, whereas the relative levels of IL-17 displayed no significant difference between OLP and normal tissues. The potential roles of IL-17 in OLP need to be elucidated further.
