Application of buccal micronucleus cytome assay in oral lichen planus and its therapeutic evaluation

Abstract:

Objective

The aim of this study was to determine the recent DNA injury and the therapeutic effect of medicine in oral lichen planus using buccal micronucleus cytome assay.

Methods

Total 39 patients with a clinicopathological diagnosis of oral lichen planus (including three clinical forms:reticular, erythematous, ulcerative) are given 4-weeks treatment:topical retinoids for reticular forms, Hydroxychloroquine and topical tacrolimus gargle for erythematous forms and intralesional triamcinolone acetonide (TA) injection and topical tacrolimus gargle for ulcerative forms. Then buccal micronucleus cytome assay and REU scoring before and after the treatment were used to determine the therapeutic effect of medicine in oral lichen planus.

Results

MEC frequency in the three forms of OLP lesions was significantly elevated (P<0.01) compared to that in adjacent normal mucosa. REU scoring in the three forms of OLP lesions was reduced (P<0.01) after clinic treatments. Only MEC frequency in ulcerative forms of OLP lesions treated with Intralesional TA injection was significantly reduced (P<0.01).

Conclusions

Medicine treatment can release the symptom of the patients with OLP. Intralesional TA injection can reduce DNA injury in oral lichen planus, and to reduce the risk of developing into cancer.
