Inhibition of gene expression of S100A4 in human periodontal ligament cells by RNA interference technology

Abstract:

Objective

Observation of using RNA interference technology by periodontal ligament(PDL) cells calcium binding protein S100A4 expression, to obtain stable cell monoclonal, for follow-up tests provide cellular level platform.

Methods

Used the technology of RNA interference (RNAi) to cut down S100A4 expression, targeting the coding sequence of human S100A4 gene and a negative control were designed. Then they were ligated into the vector of pGenesil-1 respectively,and were transfected into PDL cells. By the means of RT-PCR and Western blot,we could examine their suppression.

Result

The results showed that the interference effects of the RNAi expression vectors were 67.83%.

Conclusion

RNA interference can be effectively suppressed in periodontal ligament cells and S100A4 gene expression. This study provides a new means for investigating the effects of S100A4 in PDL cells.
