Abstract:

Objective

To study the biocompatibility of poly (hydroxybutyratehydroxyvalerate) (PHBV), a novel biopolymer membrane, and evaluate its efficiency of promoting bone regeneration in vivo.

Methods

Cell relative growth rate (RGR) was measured by MTT method to quantitatively evaluate the cytotoxicity of the biomaterials. Eight healthy dogs were used as experimental animals. Four bone defects were made on bilateral tibias in each dog.The PHBV-membrane-guided bone generation effect was assessed by histological observation.

Results

The toxicity gradation of PHBV membrane ranked from grade 0 to grade 1; new bone regeneration was observed in the defects covered with PHBV membranes at 2nd week post-surgery.The defects covered with PHBV membranes were filled with mature bone at 12th week postsurgery. The bone rehabilitation in experimental groups was earlier and better than that in control group.

Conclusion

PHBV membrane, which has no cytotoxicity to MSCs in a wide range of extract concentration, may be applied as a promising biopolymer membrane for guided bone regeneration.
