Abstract:
Objective
To investigate the mRNA and protein expressions of HIF-1 alpha and COX-2 in the rat tissues with radiation induced mucositis (ROM), and to analyze the correlation between the expression level of HIF-1 alpha and COX-2.
Methods
The rat model with ROM was established. The left buccal mucosa was excised as experimental tissue, whereas the right buccal mucosa was excised to serve as control tissue. The relative mRNA expressions of HIF-1 alpha and COX-2 were measured using the semi quantitative RT-PCR. The immunohistochemistry of Strept Avidin-Biotin-Complex Method (SABC Method) was employed to detect the distribution and intensity of HIF-1 alpha and COX-2 proteins.
Results
We successfully established a Sprague-Dawley rat model of ROM. The results of RT-PCR indicated that the left buccal mucosa expressed HIF-1 alpha and COX-2 mRNAs, while the right buccal mucosa showed negative expression of both genes. Immunohistochemical analysis demonstrated simultaneous expressions of HIF-1 alpha and COX-2 proteins in the left side mucosa.
Conclusions
The mucosa of ROM expresses HIF-1 alpha and COX-2 at mRNA and protein levels, which is correlated with the severity of ROM. A high correlation between HIF-1 alpha and COX-2 expressions has also been confirmed in this study.
