Expression of minichromosome maintenance protein7 and cell division cycle protein6 and its significance in oral squamous cell carcinoma and precancerous lesion

Abstract:

Objective

To evaluate the expression of minichromosome maintenance protein7 (Mcm7) and cell division cycle protein 6 (Cdc6) and to determine its significance in oral squamous cell carcinoma (OSCC) and precancerous lesion.

Methods

RT-PCR and immunohistochemistry analysis were performed on 47 frozen samples and 98 paraffin-embedded samples to evaluate mRNA and protein expressions of Mcm7 and Cdc6.

Results

RT-PCR results indicated positive expressions of Mcm7 mRNA in normal oral mucosa, precancerous lesions, and squamous cell carcinomas and significant differences were found among different groups (P <0.01).Cdc6 mRNA had low expression in normal oral mucosa and was highly expressed in precancerous lesions and squamous cell carcinoma, with significant differences delete among all groups (P <0.01).Immunohistochemistry analysis results indicated positive protein expression of Mcm7 in the normal oral mucosa, precancerous lesions, and squamous cell carcinomas, with a labelling index of 3.6%, 22.3% and 45.9% respectively.Significant differences were found among all groups (P <0.01).Mcm7 positive protein expression was significantly higher in the squamous cell carcinoma group than that of the precancerous lesion group (P <0.01).Cdc6 positive protein expression was lower than Mcm7 expression in all samples, with low expressions observed in normal oral mucosa,and high expressions in precancerous lesions and squamous cell carcinomas.Labelling index values among groups were statistically significant (P <0.01).Mcm7 and Cdc6 expressions in the lymph node metastasis group were significantly higher than those of the non-metastasis group (P <0.01).

Conclusions

The expression of Mcm7 and Cdc6 is positively correlated to OSCC occurrence and lymph node metastasis.Detection of the expression of Mcm7 and Cdc6 may be used for early diagnosis and prognostic judgment of oral squamous cell carcinoma.
