Expression of VEGF in the periodontium of rats with estrogen difficiency

Abstract:

Objective

To observe the expression of VEGF in the rats with estrogen difficiency,discuss the effect of estrogen on alveolar bone absorption in periodontitis rats, and investigate the effect of estrogen upon the susceptibility of periodontitis.

Methods

26 six-month-old Sprague-Dawley(SD)rats were randomly divided into three groups:group A(control), B(ovariectomy) and C(ovariectomy plus estradiol treatment). All rats were placed ligature at the upper first molars to induce periodontitis.The periodontal bone loss (PBL) and periodontal bone support (PBS) were measured on digital radiographs of the maxilla. Immunohistochemistry were used to observe the expression of VEGF in periodontium.

Results

The PBL of group OVX was higher than that of the other groups, while the PBS was lower, there was no difference between group A and group C. There was a significant difference in PBL but not in PBS between the rats sacrificed 6 weeks and 8 weeks after ovariectomy.VEGF mostly expressed in the cytoplasm of gingival epithelium cells, gingival fibroblasts, periodontal ligament fibroblasts, osteoblasts, osteoclasts, endothelial cells, monocytes, polyneuclear huge cells,and bone marrow monocytes, as well as stromal cells.

Conclusion

It is suggested that VEGF plays an important role in the pathologic process of alveolar bone resorption in rats with estrogen-deficiency.
