Detection of Th17/Treg in peripheral blood and gingival tissues of experimental rats with periodontitis

Abstract:

Objective

To observe the presentation of Th17 and Treg cells in peripheral blood and gingival tissues of SD rats with experimental periodontitis.

Methods

Experimental periodontitis of SD rats was established by silk thread ligation and P.gingivalis daubing. Th17 and Treg cells in peripheral blood were detected by the flow cytometry. PCR was used to measure the presentation of RORγt、Foxp3 mRNA in the gingival tissues. The concentrations of IL-17, IL-10 and TGF-β in peripheral blood and gingival crevicular fluid were measured using ELISA method.

Results

Th17 (F = 30.88, P = 0.00) and Treg cells (F = 147.03, P = 0.00) and the ratio of Th17/Treg (F = 219.53, P = 0.00) in the group of peripheral blood were increased compared with the control group. Presentation of RORγt (F = 35.61, P = 0.04) and Foxp3 (F = 216.60, P = 0.01) mRNA in the inflamed gingival tissues were increased compared with the control group, however, no significant difference of the ratio of RORγt/Foxp3 mRNA existed between two groups (F = 0.63, P = 0.44) . The concentrations of IL-10 in peripheral blood (F = 6.41, P = 0.02) and IL-17 (F = 9.03, P = 0.01) in gingival crevicular fluid of the experimental group significantly increased, and there were no differences of the levels of IL-17 (F = 0.08, P = 0.78) , TGF-β (F = 3.48, P = 0.06) in peripheral blood and IL-10 (F=5.85, P=0.08) in gingival crevicular fluid between two groups.

Conclusion

Expression of Th17 and Treg cells both in peripheral blood and in inflamed gingival tissues of rats with periodontitis were up-regulated, which may be associated with the progression of periodontitis, as well as some related systemic diseases.
