Rapamycin induced autophagy and suppression in tongue squamous cell carcinoma cell proliferation and migration

Abstract:

Objective

To investigate the effect of autophagy induced by rapamycin on proliferation, migration of tongue squamous cell carcinoma (TSCC) cell line.

Methods

TSCC cell line (Tca8113) was treated with rapamycin to induce autophagy.Then, Western blot was performed to examine the expression of Beclin1 and LC3.MTT assay was used to evaluate cell proliferation.Wound healing was carried out to assess cell migration.

Results

After 6 h of Rapamycin treatment, the expression of LC3 was gradually increased in Tca8113 cell.After treatment with rapamycin for 24 h, LC3-Ⅱ was dramatically increased at protein level and autophagical activity was significantly increased (P < 0.05).MTT assay indicated that cell proliferation was inhibited by Rapamycin.Migration ability was also reduced after treatment with rapamycin (P <0.05).

Conclusion

Rapamycin can induce autophagy and suppress proliferation and migration in tongue squamous cell line.
