Abstract:
Objective
To investigate the expression of HIF-1α in mandibular distraction osteogenesis using TiNi-SMA distractor and to determine the possible effects of HIF-1α on angiogenesis of distracted side.
Methods
Twelve Beagle dogs were enrolled. A 1.5 cm×1.0 cm bone defect was created in both sides of mandible in ten dogs and same shape transport bone disc was created mesially by corticotomy. TiNi-SMA distractor was settled to perform bone transport to repair defect. Two dogs were sacrified on 1, 4, 7, 10 and 15 days after operation seperatively and specimen were harvested. Two dogs were used as control without bone distraction but corticotomy was underwent in same shape in mandible and sacrified 10 days after operation. RT-PCR was employed to investigate the temporal mRNA expression levels of HIF-1α. Immunohistochemical analysis was used to verified the expression of HIF-1α and VEGF proteins. Microvessel density was also determined by detecting CD34 expression. The results were analysed by SPSS 11.5 statistical software.
Results
Positive expression of HIF-1α mRNA were detected 4,7,10 and 15 days after operation in distracted area by RT-PCR and immunohistochemical analysis respectively. Both was expressed mainly in active osteoblasts and peaked on 7th day postoperatively. VEGF expressed positively 4,7,10 and 15 days after operation and expressed both in active osteoblasts and vascular endothelial cells in distracted area, with an increase from 4th day to 10th days after operation (peaked on day 10). The values of microvessel density also increased during distraction period and peaked on the 7th day postoperatively (P <0.05). There were no apparent expression of HIF-1α mRNA, HIF-1α protein and VEGF in control group.
Conclusion
HIF-1α was highly expressed in the period of constant mandibular distraction osteogenesis. It plays an important role in the end period of distraction and may accelerate angiogenesis in distracted side by upregulating the expression of VEGF.
