目的

探讨全反式维甲酸(ATRA)对大鼠骨髓间充质细胞(BMSCs)脂肪化的诱导作用。

方法

分离大鼠BMSCs,将原代细胞分别应用普通培养基(CM)、成骨诱导培养基(OM)、ATRA、OM+ATRA 诱导培养。检测BMSCs 的增殖情况;应用脂滴油红O 染色法检测BMSCs 的分化程度;RT-PCR 检测骨形成蛋白受体(BMP-R)的表达情况。

结果

BMSCs 在4 种培养基中均生长良好。脂滴油红O 染色显示ATRA 组阳性,CM 组和OM 组阴性。培养9 d 后,ATRA 组比CM 组BMP-RⅠA 表达水平显著升高(P <0.05);OM 或ATRA 组Smad5 mRNA表达明显高于CM 组(P <0.01)。

结论

ATRA 可诱导BMSC 脂肪化及抑制骨形成。其机理可能与BMP 信号通路相关,即诱导BMP-RⅠA 的表达。

Objective

To study the induction effects of all-trans-retinoic acid(ATRA) on the adipogenic differentiation of rat bone marrow stromal cells (BMSCs).

Methods

The BMSCs were harvested from rats and then respectively treated with control medium (CM), osteogenic medium (OM), ATRA, and OM with ATRA.Proliferation of BMSCs was accessed, and Adipogenic differentiation was evaluated by Oil Red O stain; besides, the expression of BMP-R was monitored by RT-PCR.

Results

There was a significant increase of cell numbers in all groups.Cells induced by ATRA were Oil Red O positive, while the cells induced by OM or CM were negative.RT-PCR revealed that BMP-RⅠA mRNA expression in ATRA treatment group was greater than CM group (P <0.05) and Smad5 mRNA expression in OM or ATRA group was significantly higher than that CM group (P <0.01) after 9-day.

Conclusions

ATRA is an effective inducer of adipogenesis but may inhibit osteogenesis of BMSCs.The mechanism may relate to BMP signaling pathway, i.e.the expression of BMP-RⅠA induction.
