目的
探讨口腔变形链球菌(S.mutans)密度感应系统信号蛋白的体外合成及其生物活性。
方法
采用化学合成法口腔S.mutans 密度感应感受态刺激因子(CSP)信号蛋白,通过高效液相色谱仪纯化合成蛋白,质谱分析结构,通过扫描电镜对比观察CSP 信号蛋白对S.mutans 生物膜形成的影响,分析CSP 信号蛋白的生物活性。
结果
化学合成纯化S.mutans CSP 蛋白分子,加入CSP 信号肽后,S.mutans 生物膜呈团状密集分布,细菌之间存在厚实的黏性胞外分泌物,细菌黏连呈链团状。
结论
成功合成口腔S.mutans CSP 信号蛋白,该蛋白肽具备促进S.mutans 生物膜形成的生物活性。
Objective
To synthesize and analyze the biological activities of the competence stimulated peptide (CSP) in Streptococcus mutans (S.mutans) quorum sensing system.
Methods
The CSP peptide derived from S.mutans UA159 comC gene N-terminal 26-46 was chemically synthesized and purified. The biological activity of CSP, which was used as a quorum-sensing signal in S.mutans biofilm formation CSP, was analyzed by scanning electron microscope (SEM).
Results
The CSP was accurately synthesized, and SEM confirmed the addition of chemically synthetic CSP induced the adhesion of S.mutans cells and significantly increased the biofilm formation of S.mutans.
Conclusion
The CSP was chemically synthesized with biological activities, which made it possible to study the mechanism of quorum sensing system in S.mutans.
