目的

探讨人唾液腺腺样囊性癌(SACC)中转化生长因子β1(TGF-β1)、丝裂原活化蛋白激酶[MAPK(p-ERK1/2、p-p38 及p-JNK1/2)]的表达、相关性及其与临床病理特征的关系。

方法

应用免疫组织化学SP 二步法检测27 例SACC 组织和15 例正常唾液腺组织标本中TGF-β1、MAPK 的表达情况。 所有数据采用SPSS 17.0 统计软件包进行统计学分析。

结果

SACC 中TGF-β1、MAPK 的阳性表达率明显高于正常对照唾液腺(分别为P <0.01,P <0.01,P <0.01 和P <0.05)。 SACC组织中TGF-β1、p-ERK1/2 和p-p38 在Ⅲ~Ⅳ期组的阳性表达率明显高于Ⅰ~Ⅱ期组(分别为P <0.01,P <0.05 和P <0.05),但p-JNK1/2 的表达与临床分期无统计学关系(P >0.05),TGF-β1、MAPK 的表达与患者的其他临床病理因素均无统计学关系(均为P >0.05);TGF-β1 与MAPK 的表达显著正相关 (分别为r=0.819、P <0.001;r=0.728,P <0.001;r=0.640,P <0.05)。

结论

TGF-β1、MAPK 的高表达与腺样囊性癌的临床进展密切相关。 TGF-β1 可能通过激活MAPK 信号通路调控腺样囊性癌的发生、发展、侵袭及转移过程。

Objective

To investigate the expression of TGF-β1, MAPK (p-ERK1/2, p-p38 and p-JNK) in human salivary adenoid cystic carcinoma (SACC) tissues, and to explore their association and relation with clinical-pathological features.

Methods

SP immunochemistry staining method was applied to detect protein expression of TGF-β1, MAPK in the paraffin-embedded specimens from 27 cases of human SACC and 15 cases of normal human salivary epithelial tissues. All data were analyzed by SPSS 17.0 software package.

Results

The positive expression of TGF-β1, MAPK in SACC tissues increased compared with that in normal salivary tissues (P <0.01, P <0.01, P <0.01 and P <0.05 respectively). Positive expression rates of TGF-β1, p-ERK1/2 and p-p38 at the third and forth phases was higher than that at the first and second phases in SACC (P <0.01, P <0.05 and P <0.05 respectively), except p-JNK (P >0.05); their expression was not obviously correlated with patient's other clinical-pathological features (P >0.05); TGF-β1 had a positive relationship with MAPK in protein expression in SACC (r=0.819, P <0.001, r=0.728, P <0.001 and r=0.640, P <0.05 respectively).

Conclusions

Increased expression of TGF-β1, MAPK may be associated with clinical progression of SACC, and they may participate in the process of SACC initiation, progression, invasion and metastasis by TGF-β1 activated MAPK.
