目的

探讨苹果多酚(AP)体外抑制人腺样囊性癌肺高转移细胞株(ACC-M)细胞增殖的作用机制。

方法

MTT 法、流式细胞术观察不同浓度的AP 对ACC-M 细胞增殖的影响;反转录聚合酶链反应(RT-PCR)和Western blot 检测AP 作用下ACC-M 细胞血管内皮生长因子受体2(VEGFR2) mRNA 和蛋白表达的变化。 SPSS 13.0 统计学软件进行数据分析。

结果

随着AP 浓度的增加,ACC-M 细胞增殖减少,ACC-M 细胞中VEGFR2 mRNA 和蛋白表达量减少,差异有统计学意义。

结论

AP 可体外抑制ACC-M 细胞增殖,下调VEGFR2 mRNA及蛋白的表达可能是其机制之一。

Objective

To investigate the mechanism of apple polyphenols inhibiting ACCM cell proliferation in in vitro.

Methods

The effects of apple polyphenols on the proliferation in ACC-M cell was observed by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) assay and flow cytometry.Eeffects of apple polyphenols on the mRNA expression of VEGFR2 in ACC-M cell was evaluated by RT-PCR.Western blot was used to observed the influences of apple polyphenols on the protein of VEGFR2 in ACC-M cell.The data were analyzed by SPSS 13.0 software package.

Results

Compared with the control group, proliferation of ACCM cell in experimental group decreased and VEGFR2 mRNA and protein expression was down regulated significantly with the increase in the concentration of apple polyphenol.

Conclusions

Apple polyphenols could inhibit cell proliferation in ACC-M cell in vitro, and one of the mechanisms was probably to restrain the expression of the mRNA and protein of VEGFR2.
