目的
从牙石中分离Nanobacteria 菌,并进行培养和鉴定。
方法
收集15 颗牙石,随即进行Nanobacteria 菌的分离、 培养, 观察该菌形态学及生长特性, 采用von Kossa 染色、Nanobacteria 菌单克隆抗体8D10 和G1B8 免疫组化和间接免疫荧光染色、Hoechst 染色和von Kossa染色双重染色法联合进行Nanobacteria 菌鉴定。
结果
本次实验15 颗牙石中所分离、培养出的细菌形态学特征与生长特性与Nanobacteria 菌相似,von Kossa 染色呈阳性反应,单克隆抗体免疫组化染色和间接免疫荧光染色阳性。 双重染色实验中, 样本Hoechst 染色呈阴性反应,von Kossa 染色呈阳性反应, 而牙髓细胞的Hoechst 染色可见胞核呈明显阳性反应。
结论
牙石中可分离得到Nanobacteria 菌,该菌的生物学特性可能在牙石的形成中起到重要的作用。
Objective
To isolate and identify Nanobacteria from dental calculus.
Methods
Freshly collected 15 dental calculus were used for the isolation and culture of Nanobacteria. The morphology and growth characteristics of the cultured bacteria were examined. Nanobacteria was identified by von Kossa stain, immunohistochemical and immunofluorescence stain with 8D10 and G1B8 antibodies, as well as double stain including Hoechst stain and von Kossa stain.
Results
The morphology and growth characteristics of the bacteria isolated from 15 dental calculus were similar to that of Nanobacteria. Von Kossa stain, immunohistochemical stain, and immunofluorescence stain with nanobacteria-specific monoclonal antibodies were positive. In double staining test, Hoechst staining was negative while von Kossa staining was positive. In contrast, Hoechst staining of the dental pulp cells was positive.
Conclusion
The bacteria isolated from dental calculus in this study has been shown to be Nanobacteria. The unique biological characteristics of Nanobacteria may play an important role in the formation of dental calculus.
